Role of Oxygen-Derived Free Radicals in Ischemia-Reperfusion Injury to the Liver


Abstract
Ischemia/reperfusion injury of the liver is an important clinical problem, e.q., hepatic failure after shock, liver transplantation, or liver surgery. There is a rapidly growing body of evidence which indicates that oxygen free radicals play a
major
role in producing parenchymal and microvascular damages associated with reperfusion of ischemic tissues. Two enzymes are frequently invoked to explain oxygen free radical production after ischemia/reperfusion, i.e., xanthine oxidase and
neutrophilic
NADPH oxidase. The primary objective of this review is to summarize the studies that implicate oxygen radicals and xanthine oxidase (as their source) in ischemia/reperfusion injury to the liver. Evidence supporting a role for oxygen radicals in
ischemia/rperfusion injury in other organs(small intestine, stomach, pancreas and skin) is also presented.
